I've seen many comments revolving around the .45 caliber and that "it's so weak (needing two headshots for a kill) that it may as well be a .22".
While I understand your point, assuming that a .22 caliber head shot will not kill you is ignorant. As someone raised around firearms my whole life from Air rifles to assault rifles Almost everything above an air rifle is all but guaranteed to kill you with a headshot. Sure, there are exceptions, there always is. But in generalization (as a game has) you would be dead.
I say as a generalization because games on average are not designed to take random factors into account. Factors like where it hit the head, trajectory on entry and exit. The basics.
While a .22 is weak as hell compared to a .45 we are talking about a headshot. I have personally destroyed refrigerators with a .22 rifle and .22 revolver (my old favorite model 666 HK), going in the front and out the back of a closed fridge and I'll go ahead and say your skull is no match.
Not to mention, I was a Juror on a murder trial involving a man shooting another in the head with a .22. The medical examiner explained that he would rather see any other gun used in a headshot for one simple reason and I quote "If you're lucky it is fired straight and enters the victim and exits. 90% of the time the shot enters but loses enough momentum that it cannot exit, instead ricocheting off the inside of the skull several times pureeing the brain."
I really just am trying to say that firearms are deadly, no matter the caliber. End of story.
TL:DR; .22 caliber is just as deadly as any other firearm if properly/improperly used.
